In the 2011 season as a true freshman, Smith appeared in 8 games, recording a forced fumble in a 31-3 win over Ole Miss in the Egg Bowl. Preston appeared in all 13 games and led the Bulldogs with 4.5 sacks in 2012. Preston recorded 2.5 sacks, including 2.0 in a 59–26 loss to LSU as a junior in 2013.

Preston earned Southeastern Conference Defensive Lineman of the Week honors in each of the first three weeks after starting the 2014 season with a bang. Preston was projected by NFL Draft Scout and CBS as a fourth-round pick in the 2015 NFL Draft at the end of his senior season.

In the 2015 NFL Draft, Preston was selected by the Washington Redskins in the second round with the 38th overall pick. In 2015 Smith was the seventh defensive end in the draft. Preston was signed by the Redskins on May 11, 2015, to a four-year, $5.77 million contract that includes a $2.46 million signing bonus and $3.16 million guaranteed.

During a 35–18 loss to the Green Bay Packers in the NFC Wildcard Game on January 10, 2016, Preston appeared in his first career playoff game and recorded three combined tackles, a sack and a first down safety. In the NFL, Preston was voted one of the best players under the age of 25in March 2016.

Smith entered training camp as the backup outside linebacker behind Ryan Kerrigan and Junior Galette. After Junior Galette dealt with a torn Achilles during the 2016 preseason, head coach Jay Gruden named him the starting right outside linebacker. In November 2016 in a game against the Minnesota Vikings, Preston had two sacks, along with his first career interception, helping the Redskins to a 26–20 victory.

Preston sacked Mitchell Trubisky 1.5 times and had 5 tackles in a 10–3 win in his Week 1 Packer debut against the Bears. Preston sacked Joe Flacco 3 times and forced a fumble recovered by the Packers in a 27–16 win, earning him NFC Defensive Player of the Week in Week 3 against the Denver Broncos. Preston signed with the Packers on March 14, 2022, a four-year, $52.5 million extension through the 2026 season.
